Turns out something had changed the wp_user_roles value, reverting this to a value that was on an older copy of the database thankfully fixed the issue.

Thanks @VijayHardaha, so the problem seemed to be that instead of that serialized list of roles/capabilities, the value was just set to "AOK." Luckily I have another WordPress site which is working normally, so I copied the value from there and Ta-Da!
